Bullet Blender® homogenization protocol

Notes on the protocol: This protocol does not specify a particular buffer - you may choose which is most appropriate for your downstream application (nucleic acid isolation, protein extraction, etc.).

Reagents

Homogenization buffer
1 x volume of sample

Bead choices

  • PINK bead lysis kit (PINK) (for samples up to 50 mg.)
  • RED bead lysis kit (RED) (for samples between 50 and 200 mg.)
  • 0.2 mm stainless steel beads (SSB02) Use a volume of beads equivalent to 1 x the volume of the sample

Procedure

  1. Place the sample in the tube with the beads.
  2. Add a volume of buffer equal to the volume of the sample. Sample volume may be approximated by sample weight. E.g., for a 100 mg. sample, add 0.1 ml. buffer.
  3. Close the tubes tightly and place them in the Bullet Blender.
  4. Set the controls for Speed 10 and Time 5. Press Start.
  5. After the run, remove the tubes from the instrument. If a thinner homogenate is desired, you may add additional buffer to the tube.
  6. (Optional) Return the tubes to the Bullet Blender.
  7. (Optional) Set the controls for Speed 8 and Time 2. Press Start.
  8. Proceed with your downstream application.
